武夷岩茶大红袍的组织培养与快速繁殖

摘 要:

以武夷岩茶大红袍的无菌种胚为外植体, 通过对其胚的诱导、继代增殖及生根诱导培养的筛选, 建立了完整的组织培养的体系。结果表明: 武夷岩茶大红袍生长最简单的诱导培养基为MS+蔗糖30 g·L-1, 诱导率100%; 继代最适培养基为MS+TDZ 0.1 mg·L-1+蔗糖20 g·L-1, 平均增殖系数5.64; 培养基中同时添加2.5 mg·L-1 6-BA和1.5 mg·L-1 IBA壮苗效果明显; 最佳生根培养基为1/3MS+NAA 2 mg·L-1+IAA 10 mg·L-1+抗坏血酸50 g·L-1+柠檬酸50 g·L-1+蔗糖10 g·L-1, 生根率达80%以上。

Tissue culture and rapid propagation of Camellia sinensis Dahongpao

Abstract:

The aseptic embryos of Camellia sinensis Dahongpao were used as explants, and the whole tissue culture system was established through the selection of embryo induction, subculture and root induction culture. The results showed that the optimum induction medium was MS+sucrose 30 g·L-1 and the induction rate was 100%. The optimal medium for subculture was MS+TDZ 0.1 mg·L-1+sucrose 20 g·L-1, the average increment coefficient was 5.64. The effect of 2.5 mg·L-1 6-BA and 1.5 mg·L-1 IBA on seedling growth was obvious. The optimal rooting medium was 1/3MS+NAA 2.0 mg·L-1+IAA 10.0 mg·L-1+ascorbic acid 50 g·L-1+citric acid 50 g·L-1+sucrose 10 g·L-1+agar 5.5 g·L-1, the rooting rate was more than 80%.
